The Department of Transportation in Washington DC sets rules and safety standards that commerical carriers and drivers must follow.  Each state also has a DOT office which enforces the rules and regulations set at the Federal level.  Each state can also change certain rules and regulations from Federal guidelines as they see fit (such as bridge weight limits, etc.).
